Biological Background: ACSS2 Function and Localization
- ACSS2, also known as acetyl-CoA synthetase cytoplasmic (AceCS1), ACS, or ACAS2, is a member of the acyl-CoA synthetase short-chain family.
- It catalyzes the ATP-dependent ligation of acetate and coenzyme A to form acetyl-CoA, a central metabolite for lipid synthesis and energy production. Related references: PMID:10843999 PMID:28003429 PMID:38369012
- ACSS2 can also utilize propionate to form propionyl-CoA, though with lower efficiency. Related references: PMID:38369012
- The enzyme additionally converts lactate into lactoyl-CoA, linking glycolysis to novel acyl-CoA pools. Related references: PMID:39561764
- In the nucleus, ACSS2 functions as a transcriptional co-activator: upon glucose deprivation, AMPK-mediated phosphorylation triggers its nuclear translocation, where it binds TFEB and locally generates acetyl-CoA for histone H3 acetylation at lysosomal and autophagy gene promoters, supporting tumor cell survival.
