Biological Background: ACSS2 Function and Localization

  • ACSS2, also known as acetyl-CoA synthetase cytoplasmic (AceCS1), ACS, or ACAS2, is a member of the acyl-CoA synthetase short-chain family.
  • It catalyzes the ATP-dependent ligation of acetate and coenzyme A to form acetyl-CoA, a central metabolite for lipid synthesis and energy production. Related references: PMID:10843999 PMID:28003429 PMID:38369012
  • ACSS2 can also utilize propionate to form propionyl-CoA, though with lower efficiency. Related references: PMID:38369012
  • The enzyme additionally converts lactate into lactoyl-CoA, linking glycolysis to novel acyl-CoA pools. Related references: PMID:39561764
  • In the nucleus, ACSS2 functions as a transcriptional co-activator: upon glucose deprivation, AMPK-mediated phosphorylation triggers its nuclear translocation, where it binds TFEB and locally generates acetyl-CoA for histone H3 acetylation at lysosomal and autophagy gene promoters, supporting tumor cell survival. Related references: PMID:28552616
  • Beyond metabolism, ACSS2 is recruited to chromatin at memory-related neuronal genes to sustain local acetyl-CoA production for histone acetylation, a process critical for maintaining long-term spatial memory (By similarity).
  • ACSS2 is predominantly cytosolic but shuttles to the nucleus under specific signals, and its activity is modulated by post-translational modifications including acetylation and phosphorylation.

Experimental Guidance and Technical Tips

  • The immunogen spans the C-terminal region (aa 462-701) of human ACSS2; the resulting polyclonal antibody recognizes multiple epitopes, providing robust signal detection.
  • For Western blot, consider using whole-cell lysates from human, mouse, or rat cell lines; a specific band is expected around 79 kDa.
  • In immunofluorescence/ICC, ACSS2 may localize to both cytoplasm and nucleus depending on cell state; optimize fixation and permeabilization conditions to preserve both compartments.
  • For ELISA, perform titration to determine optimal coating antigen concentration and antibody dilution.
  • Validate the antibody in your specific experimental system, as reactivity and background may vary.
