Biological Background: ACSS2 Function and Localization
- ACSS2 encodes cytoplasmic acetyl-coenzyme A synthetase, also known as acetate--CoA ligase, AceCS1, or acyl-CoA synthetase short-chain family member 2, and belongs to the acyl-CoA synthetase short-chain family.
- Catalyzes the synthesis of acetyl-CoA from short-chain fatty acids, with acetate as the preferred substrate; can also utilize propionate with lower affinity, and converts lactate into lactoyl-CoA. Related references: PMID:10843999, PMID:28003429, PMID:38369012, PMID:39561764
- Nuclear ACSS2 promotes lysosomal biogenesis and autophagy via AMPK-mediated phosphorylation and translocation, where it binds TFEB and provides acetyl-CoA for histone acetylation at target gene promoters, supporting tumor survival. Related references: PMID:28552616
- Localizes to both cytoplasm and nucleus, enabling distinct metabolic and gene-regulatory roles.
- Functions as a chromatin-bound transcriptional coactivator that maintains local acetyl-CoA pools for histone acetylation, linking metabolism to epigenetic regulation and long-term spatial memory.
- Subject to phosphorylation (by AMPK) and acetylation, which modulate subcellular localization and activity.
- Alternative splicing generates multiple isoforms; proteomics confirms expression across tissues, with high levels in liver, kidney, and heart.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 50–150 of human ACSS2; consider using this region to verify antibody specificity in your assay.
- For WB, validate the antibody in the relevant cell or tissue lysate, and consider nuclear/cytoplasmic fractionation to confirm localization-dependent expression.
- Test cross-reactivity in mouse and rat samples as indicated; optimize dilution and detection conditions empirically.
- In ELISA, include appropriate controls and verify linearity and sensitivity for your specific analyte range.
