Biological Background: ACSS2 Function and Localization
- ACSS2 encodes cytoplasmic acetyl-coenzyme A synthetase, a member of the acyl-CoA synthetase short-chain family, also known as AceCS1.
- Catalyzes the synthesis of acetyl-CoA from acetate (preferred substrate) and CoA, with lower affinity for propionate, and also converts lactate to lactoyl-CoA; does not act on butyrate or crotonate. Related references: PMID:10843999 PMID:28003429 PMID:28552616 PMID:38369012 PMID:39561764
- Central to lipid metabolism and energy homeostasis, linking acetate metabolism to acetyl-CoA pools for fatty acid and cholesterol synthesis.
- Under glucose deprivation, AMPK-mediated phosphorylation triggers nuclear translocation of ACSS2, where it binds TFEB and locally produces acetyl-CoA for histone acetylation, promoting lysosomal biogenesis, autophagy and tumorigenesis. Related references: PMID:28552616
- Acts as a chromatin-bound transcriptional coactivator for memory-related neuronal genes, maintaining local acetyl-CoA pools to support histone acetylation and gene expression (By similarity).
