Biological Background: ACSL5 Function and Localization
- ACSL5 (encoded by the ACSL5 gene) is a long-chain-fatty-acid--CoA ligase 5, also known as arachidonate--CoA ligase or LACS 5.
- Catalyzes the conversion of long-chain fatty acids into their active CoA derivatives for incorporation into cellular lipids or degradation via beta-oxidation. Related references: PMID:17681178 PMID:22633490 PMID:24269233
- Activates exogenous fatty acids for triacylglycerol synthesis and intracellular storage.
- Preferentially utilizes a wide range of saturated fatty acids, with a higher preference for C16-C18 unsaturated fatty acids.
- Localizes to the mitochondrion, endoplasmic reticulum, and cell membrane; specifically associates with the mitochondrial outer membrane and ER membrane.
- At the villus tip of the small intestine, ACSL5 may sensitize epithelial cells to TRAIL-induced apoptosis.
- May play a role in glioma cell survival, suggesting relevance in cancer biology.
- Post-translational modifications include acetylation and phosphorylation; alternative splicing generates multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (amino acids 500–739) of human ACSL5; verify recognition if your target includes N-terminal modifications or variants.
- For Western blot, anticipate a band at ~76 kDa; consider running a positive control (e.g., ACSL5-transfected cell lysate) for validation.
- The antibody cross-reacts with human, mouse, and rat; confirm performance in your specific sample type and species.
- For ELISA, the recombinant immunogen protein can serve as a positive control or coating antigen during assay optimization.
