Biological Background: ACSL3 Function and Localization
- ACSL3 is a long-chain-fatty-acid--CoA ligase that activates long-chain fatty acids for cellular lipid synthesis and beta-oxidation. Related references: PMID:22633490
- It preferentially uses myristate, laurate, arachidonate, and eicosapentaenoate as substrates, with both isoforms showing equivalent activity.
- Required for incorporation of fatty acids into phosphatidylcholine, the major phospholipid on VLDL surfaces, playing a role in hepatic lipogenesis. Related references: PMID:18003621
- Also known as medium-chain acyl-CoA ligase Acsl3, with aliases including ACS3, FACL3, LACS3, and PRO2194.
- Localizes to mitochondrial outer membrane, peroxisome membrane, microsome membrane, and endoplasmic reticulum membrane.
- Contains a transmembrane helix and uses ATP and magnesium for enzymatic activity, classified as a ligase and phosphoprotein.
