Biological Background: ACSL1 Function and Localization
- ACSL1 encodes a long-chain-fatty-acid--CoA ligase that converts fatty acids to acyl-CoAs, supporting both lipid synthesis and beta-oxidation. It preferentially uses palmitoleate, oleate, and linoleate.
Related references: PMID:21242590, PMID:22633490, PMID:24269233 - It also activates arachidonate, showing higher preference for arachidonate than epoxyeicosatrienoic acids (EETs) or hydroxyeicosatrienoic acids (HETEs).
- The protein localizes to multiple membrane compartments, including mitochondrial outer membrane, peroxisome, microsome, and endoplasmic reticulum.
- ACSL1 is highly expressed in liver, heart, skeletal muscle, kidney, and erythroid cells, with lower levels in brain, lung, placenta, and pancreas.
- Alternative splicing generates multiple isoforms, contributing to functional diversity.
- Post-translational modifications include acetylation, nitration, phosphorylation, and glycosylation, which may regulate its activity and localization.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 46-145) corresponds to a region conserved across human, mouse, and rat, supporting cross-reactivity observed.
- For Western blot, consider testing lysates from ACSL1-rich tissues such as liver or heart, and optimize antibody concentration and detection conditions.
- In immunofluorescence, use appropriate permeabilization and blocking steps given the multi-membrane localization of ACSL1.
- For immunoprecipitation, the antibody may capture native ACSL1 from whole-cell lysates; validate with appropriate positive controls.
