Biological Background: ACSF3 Function and Localization
- ACSF3 (acyl-CoA synthetase family member 3) is a mitochondrial enzyme also known as malonate--CoA ligase.
- It catalyzes the activation of malonate and methylmalonate to their CoA thioesters, initiating intramitochondrial fatty acid synthesis. Related references: PMID:21841779 / PMID:21846720
- The enzyme may exhibit a preference for very-long-chain fatty acid substrates. Related references: PMID:17762044
- ACSF3 is localized to the mitochondrion, consistent with its role in mitochondrial fatty acid and lipid metabolism.
- The protein contains ATP-binding and nucleotide-binding domains, characteristic of the acyl-CoA synthetase/ligase family.
- UniProt keywords include ATP-binding, Fatty acid metabolism, Ligase, Lipid metabolism, Mitochondrion, Nucleotide-binding, Proteomics identification, Reference proteome, and Transit peptide.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 370-576 of human ACSF3, which covers a large portion of the C-terminal region; the resulting polyclonal antibody may recognize multiple epitopes across this region.
- For western blotting, the predicted molecular weight of ACSF3 is approximately 64 kDa; a specific band may be observed near this size, but validation in the relevant sample system is recommended.
- This antibody has been tested in WB, IHC-P, and ELISA; consider optimizing dilution and detection conditions for each application.
- Cross-reactivity with mouse ACSF3 extends the utility of this antibody to mouse model studies; however, sequence homology should be checked for other species.
