Biological Background: ACRV1 (SP-10) Function and Localization
- ACRV1 (Acrosomal Vesicle Protein 1), also known as SP-10 (Sperm Protein 10) or SPACA2, is encoded by the ACRV1 gene located on chromosome 11q13 in humans.
- The protein is classified as an acrosomal component, specifically localized to the acrosome, a cap-like secretory vesicle covering the anterior portion of the sperm nucleus.
- ACRV1 expression is highly testis-specific, consistent with its role in sperm development and acrosome biogenesis.
- According to UniProt, ACRV1 contains multiple repeat regions and a signal peptide, suggesting it is a secreted glycoprotein that undergoes post-translational processing.
- Post-translational modifications include glycosylation, and the protein is subject to alternative splicing, which may generate distinct isoforms.
- The mature protein has a molecular weight of approximately 28 kDa.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 22–265, representing a large portion of the extracellular region of human ACRV1. This design aims to recognize both full-length and processed forms of the protein.
- For Western blot analysis, a band near 28 kDa is expected in human testis tissue lysates. Given cross-reactivity with rat, rodent models may be suitable for validation studies.
- Since ACRV1 is a glycoprotein, enzymatic deglycosylation can be employed to verify band specificity and investigate glycoforms.
- The antibody is applicable in ELISA; consider using it as a capture antibody paired with a detection antibody, or as a detection antibody in indirect ELISA setups, with appropriate optimization of coating and blocking conditions.
