Biological Background: ACOX3 Function and Localization
- ACOX3, also designated branched-chain acyl-CoA oxidase (BRCACox) or pristanoyl-CoA oxidase, is encoded by the gene ACOX3 (BRCOX, PRCOX) and regulated by alternative splicing.
- ACOX3 catalyzes the rate-limiting dehydrogenation step in peroxisomal beta-oxidation of branched-chain fatty acids. It preferentially oxidizes 2-methyl-branched substrates such as pristanoyl-CoA, producing hydrogen peroxide as a byproduct, and can also act on straight-chain substrates with lower efficiency. - For Western blotting, the predicted molecular weight is approximately 78 kDa. Validate the band size in your sample lysates, considering possible alternative splicing variants.
- In immunofluorescence/immunocytochemistry (IF/ICC), expect punctate peroxisomal staining. Co-localization with a peroxisomal marker (e.g., PMP70) is recommended for confirmation.
- For ELISA, the antibody may be used for direct or sandwich detection. Optimize coating and antibody concentrations experimentally.
- Liver or kidney tissue lysates serve as suitable positive controls, given the documented protein expression in these organs.
