Biological Background: ACOX3 Function and Localization

  • ACOX3, also designated branched-chain acyl-CoA oxidase (BRCACox) or pristanoyl-CoA oxidase, is encoded by the gene ACOX3 (BRCOX, PRCOX) and regulated by alternative splicing.
  • ACOX3 catalyzes the rate-limiting dehydrogenation step in peroxisomal beta-oxidation of branched-chain fatty acids. It preferentially oxidizes 2-methyl-branched substrates such as pristanoyl-CoA, producing hydrogen peroxide as a byproduct, and can also act on straight-chain substrates with lower efficiency. (PubMed:29287774) Related references: PMID:29287774
  • The enzyme localizes exclusively to peroxisomes, consistent with its role in lipid metabolism.
  • ACOX3 is a flavoprotein that requires FAD as a cofactor and is subject to post-translational modifications including acetylation and phosphorylation.
  • At the protein level, ACOX3 is expressed in liver, kidney, lung, brain (cortex and cerebellum), and heart.
  • It belongs to the acyl-CoA oxidase family and exhibits functional redundancy with other peroxisomal oxidases in branched-chain fatty acid metabolism.

Experimental Guidance and Technical Tips

  • The immunogen used is a recombinant fusion protein spanning amino acids 10–100 of human ACOX3 (NP_001095137), an N-terminal region conserved across human, mouse, and rat, supporting the reported cross-reactivity.
  • For Western blotting, the predicted molecular weight is approximately 78 kDa. Validate the band size in your sample lysates, considering possible alternative splicing variants.
  • In immunofluorescence/immunocytochemistry (IF/ICC), expect punctate peroxisomal staining. Co-localization with a peroxisomal marker (e.g., PMP70) is recommended for confirmation.
  • For ELISA, the antibody may be used for direct or sandwich detection. Optimize coating and antibody concentrations experimentally.
  • Liver or kidney tissue lysates serve as suitable positive controls, given the documented protein expression in these organs.
