Biological Background: ACOX2 Function and Localization
- ACOX2 (peroxisomal acyl-coenzyme A oxidase 2) is also known as BCOX, BRCOX, CBAS6, THCCox, or BRCACOX.
- ACOX2 catalyzes the initial and rate-limiting dehydrogenation step in peroxisomal beta-oxidation of branched-chain fatty acids and bile acid intermediates, producing hydrogen peroxide as a by-product. Related references: PMID:29287774
- It stereospecifically oxidizes substrates with (2S)-methyl groups, such as (2S)-pristanoyl-CoA, and exhibits redundancy with ACOX3. Related references: PMID:29287774
- In the liver, ACOX2 mediates side-chain beta-oxidation of C27 bile acyl-CoA intermediates with a (25S)-methyl group, a critical step in peroxisomal bile acid synthesis. Related references: PMID:27884763 PMID:29287774
- In brown adipose tissue, ACOX2 participates in thermogenesis by beta-oxidizing monomethyl branched-chain fatty acids, coupling to AMPK activation and mitochondrial ATP production.
