Biological Background: ACOX1 Function and Localization
-
ACOX1 (peroxisomal acyl-coenzyme A oxidase 1), also known as palmitoyl-CoA oxidase or straight-chain acyl-CoA oxidase (SCOX), catalyzes the initial and rate-limiting step of peroxisomal beta-oxidation of straight-chain saturated and unsaturated very-long-chain fatty acids (VLCFAs). Related references: PMID:15060085 PMID:17458872 PMID:17603022 PMID:7876265
-
The enzyme desaturates 2,3-saturated acyl-CoA to 2-trans-enoyl-CoA, donating electrons directly to molecular oxygen to produce hydrogen peroxide. Related references: PMID:17458872 PMID:17603022 PMID:7876265
-
Exhibits broad substrate activity, with highest activity toward medium-chain fatty acyl-CoAs (optimum at C10-decanoyl-CoA) and activity extending to long-chain and VLCFA-CoAs.
-
Localized exclusively to the peroxisome, where it participates in lipid metabolism.
-
Exists as alternatively spliced isoforms (isoform 1 and isoform 2) with tissue-specific expression; isoform 1 predominates in liver and kidney, while isoform 2 is higher in brain, lung, muscle, white adipose tissue, and testis, with highest overall expression in testis.
-
Contains FAD as a cofactor and is subject to post-translational modifications including acetylation and phosphorylation, reflecting its regulated role in fatty acid oxidation.
-
May contribute to polyunsaturated fatty acid (PUFA) biosynthesis and systemic docosahexaenoic acid (DHA) homeostasis through peroxisomal beta-oxidation of tetracosahexaenoic acid (THA), with implications in inter-organ VLCFA signaling and adipose tissue thermogenesis regulation.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein covering amino acids 1–270 of human ACOX1; the antibody is expected to recognize the N-terminal region and may detect full-length and relevant isoform forms.
- For Western blot (WB), the predicted molecular weight is approximately 74 kDa; validate band specificity using appropriate controls and blocking conditions.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), consider antigen retrieval protocols and include tissue-specific positive and negative controls, especially in testis, liver, and adipose tissue.
- For ELISA applications, optimize antibody coating concentration and detection reagents according to the specific assay format.
- Due to cross-reactivity with human, mouse, and rat, the antibody can be used in comparative studies across these species; verify performance in the target sample matrix to ensure consistent results.
