- ACO1 gene encodes cytoplasmic aconitate hydratase, also known as iron regulatory protein 1 (IRP1) or IRE-BP1, a bifunctional enzyme that senses intracellular iron levels and switches between two distinct activities.
- As an aconitase, it catalyzes the isomerization of citrate to isocitrate via cis-aconitate in the cytoplasm when cellular iron is sufficient; this activity depends on the presence of a 4Fe-4S cluster.
- Under iron deprivation, ACO1 loses its iron-sulfur cluster and acquires mRNA-binding activity, interacting with iron-responsive elements (IREs) in untranslated regions of target mRNAs to post-transcriptionally regulate genes involved in iron uptake, storage, and utilization.
