Biological Background: Acetylcholinesterase Function and Localization
- Acetylcholinesterase (ACHE) is a serine esterase that catalyzes the rapid hydrolysis of the neurotransmitter acetylcholine at cholinergic synapses, terminating synaptic transmission.
- The human ACHE gene encodes multiple isoforms via alternative splicing, including the erythrocytic isoform H, commonly referred to by aliases such as YT, ACEE, ARACHE, and N-ACHE.
- ACHE belongs to the cholinesterase family and plays a critical role in neuromuscular junction signaling and neuronal apoptosis regulation.
- Subcellularly, ACHE localizes to the synapse, cell membrane, nucleus, and is also secreted; it exists as both membrane-bound (GPI-anchored) and soluble forms.
- The protein undergoes post-translational modifications including disulfide bond formation and glycosylation, and carries blood group antigen determinants.
- Isoform H is highly expressed in erythrocytes, consistent with its anchored membrane form and role in acetylcholinesterase activity in blood.
Experimental Guidance and Technical Tips
- The immunogen spans the C-terminal region (aa 398-617) of human ACHE, which may be conserved across species; consider verifying sequence homology for mouse and rat targets.
- For Western blot (WB), the predicted molecular weight is ~68 kDa; include appropriate positive controls such as erythrocyte or neuronal lysates.
- In immunofluorescence/ICC, note that ACHE can localize to the cell membrane, nucleus, and is secreted; permeabilization and fixation conditions may need optimization.
- For ELISA applications, use recombinantly expressed ACHE or native protein from serum/plasma as capture antigen; validate with known standards.
