Biological Background: ACE2 Function and Localization
- ACE2 (Angiotensin-Converting Enzyme 2), also known as ACEH, is a carboxypeptidase that critically regulates the renin-angiotensin system (RAS) by converting Angiotensin II to the vasodilator Angiotensin 1-7, and Angiotensin I to Angiotensin 1-9. Related references: PMID:10924499 PMID:10969042 PMID:11815627
- It also cleaves other vasoactive peptides including neurotensin, kinetensin, des-Arg bradykinin, apelins, casomorphins, and dynorphin A, acting as a counter-regulatory enzyme to ACE. Related references: PMID:10969042 PMID:11815627 PMID:27217402 PMID:28293165
- ACE2 serves as the cellular receptor for SARS-CoV and SARS-CoV-2, facilitating viral entry.
- Localized to the cell membrane, particularly the apical membrane of epithelial cells, as well as in the cytoplasm and secreted forms.
- Expressed in heart, kidney, testis, and gastrointestinal tract; low expression in lung alveolar type 2 cells, nasal and bronchial epithelial cells.
- ACE2 gene produces multiple isoforms through alternative splicing.
- Keywords: carboxypeptidase, metalloprotease, host cell receptor for virus entry, zinc-binding, glycoprotein, phosphoprotein (UniProt Q9BYF1).
Experimental Guidance and Technical Tips
- The immunogen covers a large portion of the extracellular domain (aa 18–740), potentially allowing recognition of both native and denatured ACE2.
- For Western blot under reducing conditions, a band around 92 kDa is expected; validate with positive controls such as kidney or heart lysates.
- For immunohistochemistry (IHC-P), consider antigen retrieval methods and validate on kidney or intestinal tissue sections.
- For immunofluorescence (IF-P), permeabilization may be necessary for intracellular epitope access; co-localization with membrane markers is advised.
- Cross-reactivity with mouse and rat ACE2 is listed; verify specificity via sequence alignment and using appropriate controls.
- In SARS-CoV-2 research, spike protein competition assays can confirm specificity for the receptor-binding domain.
