  • ACE2 is a zinc metalloprotease carboxypeptidase that critically regulates the renin-angiotensin system by converting angiotensin II to angiotensin 1-7, acting as a vasodilator and counterbalancing the effects of angiotensin II vasoconstriction. Experimental Guidance and Technical Tips

  • The immunogen spans residues 18–740 of human ACE2, encompassing the entire extracellular domain. This design is expected to detect full-length endogenous ACE2 across multiple applications.
  • For Western blot, a specific band near 92 kDa should be observed under reducing conditions. Use validated positive control lysates from human kidney, heart, or intestinal tissue, or from mouse/rat tissues given reported cross-reactivity.
  • For immunohistochemistry (paraffin-embedded) and immunofluorescence, antigen retrieval is often required. Kidney proximal tubules, small intestine enterocytes, and testis serve as strong positive controls due to high ACE2 expression.
  • In ELISA assays, this antibody may function as capture or detection reagent. Titration and pairing with a second ACE2 antibody clone are recommended for optimal sandwich ELISA performance.
