  • ACD (Adrenocortical dysplasia protein homolog), also known as POT1 and TIN2-interacting protein, is a component of the shelterin (telosome) complex that protects chromosome ends by binding to double-stranded TTAGGG repeats.
  • It promotes the binding of POT1 to single-stranded telomeric DNA, modulating the inhibitory effects of POT1 on telomere elongation.
  • The ACD-POT1 heterodimer enhances telomere elongation by recruiting telomerase to telomeres and increasing its processivity.
  • This protein localizes to the nucleus and is specifically associated with chromosome telomeres.
  • Alternative splicing produces multiple isoforms, and post-translational modifications include phosphorylation and ubiquitin-like conjugation (isopeptide bond).
  • Disease variants of ACD are associated with dyskeratosis congenita, highlighting its critical role in telomere biology.
