Biological Background: Acetyl-CoA acetyltransferase, mitochondrial Function and Localization
- Acetyl-CoA acetyltransferase, mitochondrial (EC 2.3.1.9), also known as acetoacetyl-CoA thiolase, T2, MAT, ACAT, or THIL, is encoded by the ACAT1 gene (Gene ID: 38) in humans.
- It catalyzes the last step of the mitochondrial beta-oxidation pathway, thiolytically cleaving medium- to long-chain 3-oxoacyl-CoAs into acetyl-CoA and a shortened acyl-CoA (PubMed:1715688, PubMed:7728148, PubMed:9744475).

Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 1–145 of human ACAT1 (NP_000010.1), covering the N-terminal region which includes the mitochondrial transit peptide and part of the mature protein. This design may favor detection of precursor and mature forms.
- Recommended for western blot (WB) and ELISA applications. For WB, a predominant band near 45 kDa is expected in human, mouse, and rat lysates.
- As a polyclonal antibody, potential cross-reactivity with homologous family members or ACAT1 isoforms from alternative splicing should be considered; validation in the relevant sample matrix is advised.
- For ELISA, consider using recombinant ACAT1 or mitochondrial-enriched protein extracts as coating antigen, and include appropriate controls for specificity.
