Biological Background: ACAP2 Function and Localization
- ACAP2 (Arf-GAP with coiled-coil, ANK repeat and PH domain-containing protein 2), also known as Centaurin-beta-2 (Cnt-b2), is a multidomain GTPase-activating protein.
- It functions as a GTPase-activating protein (GAP) for ADP-ribosylation factor 6 (ARF6), but does not show GAP activity for RAB35. Related references: PMID:30905672
- Contains ANK repeats, coiled-coil regions, a PH domain, and a zinc-finger motif, which mediate protein–protein interactions and membrane targeting.
- Localizes to the cell membrane and endosome membrane, consistent with its role in endosomal trafficking and remodeling of the actin cytoskeleton.
- Widely expressed in human tissues, with the highest expression level observed in the lung.
- Post-translational modifications include phosphorylation (phosphoprotein), and metal-binding via its zinc-finger domains.

- For western blot (WB) analysis, the predicted molecular weight is 88 kDa; ensure adequate separation and transfer conditions for proteins of this size.
