Biological Background: ACADS/SCAD Function and Localization
- Also known as butyryl-CoA dehydrogenase, it is a mitochondrial enzyme belonging to the acyl-CoA dehydrogenase family.
- Catalyzes the initial step of the mitochondrial fatty acid beta-oxidation pathway, converting straight-chain fatty acyl-CoA thioesters into trans-2-enoyl-CoA.
- Shows substrate specificity for saturated fatty acyl-CoA esters with chain lengths of 4 to 6 carbons. Related references: PMID:11134486 PMID:21237683
- This enzymatic reaction is FAD-dependent, serving as an electron acceptor in the dehydrogenation process.
- Localizes to the mitochondrial matrix, consistent with its role in fatty acid catabolism.
- Post-translational modifications include acetylation and phosphorylation, which may regulate enzyme activity.
- Mutations in ACADS are associated with short-chain acyl-CoA dehydrogenase deficiency, an inborn error of metabolism.
- Contains a transit peptide that directs its import into mitochondria.
Experimental Guidance and Technical Tips
- The immunogen was a synthetic peptide corresponding to amino acids 313-412 of human ACADS/SCAD (P16219). This region may determine epitope specificity.
- For Western blot, the expected molecular weight is approximately 44 kDa. Consider using appropriate reducing conditions and molecular weight markers.
- In immunohistochemistry (IHC-P), proper antigen retrieval and detection methods should be optimized for the target tissue.
- Cross-reactivity is reported with human, mouse, and rat. Validate the antibody for your specific species and sample type.
- For ELISA, employ the appropriate secondary detection system and include controls for accurate quantification.
