Biological Background: ACADM Function and Localization

  • Medium-chain specific acyl-CoA dehydrogenase (MCAD, encoded by ACADM) is a mitochondrial matrix enzyme that catalyzes the rate-limiting first step of fatty acid β-oxidation: the FAD-dependent dehydrogenation of fatty acyl-CoA thioesters to trans-2-enoyl-CoA, with electrons transferred to the electron transfer flavoprotein (ETF).   • MCAD displays substrate preference for medium-chain saturated fatty acyl-CoAs (C6–C12), but can also act on longer chains (C14, C16). Related references: PMID:1970566, PMID:21237683, PMID:2251268

  • Localized to the mitochondrial matrix, ACADM is ubiquitously expressed with highest levels in heart, liver, and skeletal muscle.

  • The immunogen was a recombinant protein covering residues 1-220 of human ACADM, suggesting the antibody may recognize multiple N-terminal epitopes. For western blot, expect a band at approximately 47 kDa; validate detection using mitochondria-rich samples such as liver or muscle lysates.

  • This antibody has been tested in WB, IF/ICC, and ELISA. For IF/ICC, mitochondrial staining patterns should be confirmed with organelle markers. Optimize dilutions empirically for each application and cell/tissue type.

  • Cross-reactivity with mouse and rat is indicated, making the antibody suitable for translational studies; however, individual verification in these species is recommended.
