Biological Background: ACADL Function and Localization
- ACADL (acyl-CoA dehydrogenase long chain) is also known as LCAD and ACAD4, a mitochondrial enzyme essential for fatty acid beta-oxidation.
- It catalyzes the first step of the mitochondrial fatty acid beta-oxidation pathway, performing the alpha,beta-dehydrogenation of long-chain fatty acyl-CoA thioesters to trans-2-enoyl-CoA using electron transfer flavoprotein (ETF) as the electron acceptor. - Each cycle of beta-oxidation shortens the fatty acid chain by two carbons, producing acetyl-CoA, FADH₂, and NADH, which fuel the citric acid cycle and oxidative phosphorylation for ATP production. Related references: PMID:17564966 PMID:26474213

- The immunogen comprises amino acids 31-210, a region that includes part of the N-terminal domain; the antibody is expected to recognize the full-length ACADL protein.
- For Western blot (WB), the observed band may be around 48 kDa (consistent with the predicted molecular weight of 47.7 kDa), but post-translational modifications could cause slight shifts.
- IHC-P may require antigen retrieval; optimization is recommended depending on tissue type and fixation methods.
- Cross-reactivity with mouse and rat suggests utility in rodent models; however, validate in the relevant sample system before use.
