Biological Background: ACAA2 Function and Localization
- ACAA2, also known as 3-ketoacyl-CoA thiolase or acetyl-CoA acetyltransferase, is a mitochondrial enzyme encoded by the ACAA2 gene.
- It catalyzes the final step of mitochondrial fatty acid β-oxidation, thiolytically cleaving medium- to long-chain 3-oxoacyl-CoAs to produce acetyl-CoA and a shortened fatty acyl-CoA.
- ACAA2 can also condense two acetyl-CoA molecules to form acetoacetyl-CoA, potentially contributing to ketone body synthesis.
- The enzyme exhibits hydrolase activity on various fatty acyl-CoAs, generating acetate in a side reaction to β-oxidation. Related references: PMID:25478839
- ACAA2 has been shown to abolish BNIP3-mediated apoptosis and mitochondrial damage. Related references: PMID:18371312
- Subcellularly, ACAA2 localizes to the mitochondrion, consistent with its role in fatty acid metabolism.
