Biological Background: ACAA2 Function and Localization
- Encodes mitochondrial 3-ketoacyl-CoA thiolase (also known as acetyl-CoA acetyltransferase, beta-ketothiolase, T1), a key enzyme in fatty acid beta-oxidation.
- Catalyzes the thiolytic cleavage of medium- to long-chain 3-oxoacyl-CoAs into acetyl-CoA and a shortened acyl-CoA, the final step of mitochondrial beta-oxidation.
- Exhibits hydrolase activity on various fatty acyl-CoAs, potentially generating acetate as a side reaction. Related references: PMID:25478839
- Also catalyzes the condensation of two acetyl-CoA molecules to acetoacetyl-CoA, suggesting involvement in ketone body metabolism.
- Abolishes BNIP3-mediated apoptosis and mitochondrial damage, indicating a role in cell survival. Related references: PMID:18371312
- Localizes to the mitochondrion.
- Post-translationally modified by acetylation and phosphorylation; contains an N-terminal transit peptide for mitochondrial import.
- Belongs to the thiolase family and is essential for energy production from fatty acids.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 298–397 near the C-terminus; verify epitope accessibility in your sample system.
- For Western blot, expect a specific band at approximately 42 kDa; use mitochondrial-enriched lysates to enhance signal.
- In immunofluorescence/ICC, the antibody should detect mitochondrial localization; consider co-staining with a mitochondrial marker.
- ELISA performance may be optimized by using the immunizing peptide as a positive control; validate dilution factors empirically.
