Biological Background: ABR Function and Localization
- ABR, also known as Active breakpoint cluster region-related protein or MDB, is a member of the breakpoint cluster region (BCR) family of proteins. It is a multidomain GTPase regulator.
- Functions as a GTPase-activating protein (GAP) for RAC1, RAC2, and CDC42, accelerating GTP hydrolysis and down-regulating their active forms. Related references: PMID:17116687 PMID:7479768
- Also possesses Dbl homology (DH) domain with guanine nucleotide exchange factor (GEF) activity, promoting conversion of CDC42, RHOA, and RAC1 from GDP- to GTP-bound state. Related references: PMID:7479768
- Acts as a negative regulator of neuronal RAC1 activity and modulates macrophage functions such as CSF-1 directed motility and phagocytosis (By similarity).
- Subcellular localization: found at dendritic spines, axons, and synapses.
- Expression is highly enriched in the brain, with weaker expression in heart, lung, and muscle.
- Classified as a phosphoprotein and expressed as multiple isoforms due to alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen peptide corresponds to the C-terminal region; thus, the antibody may recognize ABR isoforms containing this sequence. Validate in the relevant sample system.
- For Western blot, a band around 98 kDa is expected. Consider using brain tissue lysate as a positive control due to high ABR expression.
- The antibody cross-reacts with mouse and rat; it may be suitable for studies in rodent models. Confirm specificity in the species of interest before use.
- For ELISA, optimize coating and detection conditions empirically to achieve optimal performance.
