Biological Background: ABI1 Function and Localization
- ABI1 (Abl interactor 1, also known as E3B1, SSH3BP1) is an adapter protein that negatively regulates cell growth and transformation by interacting with nonreceptor tyrosine kinases ABL1 and ABL2.
- It plays a key role in cytoskeletal reorganization and signal transduction: together with EPS8 and SOS1, ABI1 forms a trimeric complex that exhibits Rac-specific guanine nucleotide exchange factor (GEF) activity, linking Ras to Rac signaling.
- ABI1 participates in EGFR signaling and modulates the EGF-induced Erk pathway; it also regulates ABL1-mediated phosphorylation of ENAH and recruits WASF1 to lamellipodia, influencing actin dynamics.
- In neurons, ABI1 promotes dendritic outgrowth and branching, and helps determine the shape and number of synaptic contacts during development.
- Subcellular localization includes the cytoplasm, nucleus, and actin-rich structures such as lamellipodia, filopodia, growth cones, and postsynaptic densities.
- The protein contains an SH3 domain and is subject to phosphorylation, acetylation, and alternative splicing; it is widely expressed with highest levels in brain.

- The immunogen (amino acids 180–230) lies within a region that is conserved across human, mouse, and rat, consistent with the antibody's reported cross-reactivity; validate reactivity in your specific species and tissue context.
- For Western blot, a major band around 55 kDa is expected; use appropriate positive and negative cell/tissue lysates and consider both reducing and non-reducing conditions.
- In immunohistochemistry (paraffin-embedded), optimize antigen retrieval (e.g., citrate buffer, pH 6.0) and antibody titration; a known positive control (e.g., brain or tumor tissue) is recommended.
- For ELISA, the antibody can serve as a capture or detection reagent; pair with a suitable matched antibody and determine optimal coating concentrations empirically.
