Biological Background: ABCG1 Function and Localization
- ABCG1 (ATP-binding cassette sub-family G member 1), also known as ABC8 and White protein homolog, is encoded by the ABCG1 gene and belongs to the ABC transporter superfamily.
- It functions as an ATP-dependent transporter that catalyzes the efflux of phospholipids (such as sphingomyelin), cholesterol, and its oxygenated derivatives like 7beta-hydroxycholesterol; this transport is ALB-dependent. Related references: PMID:17408620, PMID:24576892, PMID:16702602
- ABCG1 is an active component of the macrophage lipid export complex and prevents cell death by transporting cytotoxic 7beta-hydroxycholesterol. Related references: PMID:17408620
- The protein may also participate in intracellular lipid transport processes and helps maintain cellular lipid homeostasis in various cell types, not limited to macrophages.
- Subcellularly, ABCG1 is localized to the endoplasmic reticulum membrane, Golgi apparatus membrane, and cell membrane.
- Post-translational modifications include palmitoylation, and the protein contains characteristic transmembrane helices and nucleotide-binding domains typical of ABC transporters.
- ABCG1 is expressed in several tissues with higher levels in macrophages; its expression is upregulated in macrophages from Tangier disease patients.

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 6-120 of human ABCG1, targeting the N-terminal region. For Western blot, a band around 76 kDa is expected; validate in your sample system with appropriate loading controls.
- For immunofluorescence (IF-P) and immunohistochemistry (IHC-P), test antibody dilutions in known positive tissues or cells (e.g., macrophages or cell lines expressing ABCG1) and use appropriate fixation/permeabilization protocols.
- This polyclonal antibody is suitable for ELISA; consider pairing it with an appropriate detection antibody for sandwich ELISA applications.
- As a polyclonal reagent, lot-to-lot variability is possible; titration and inclusion of positive/negative controls are recommended for each new lot.
