Biological Background: ABCE1 Function and Localization
- ABCE1, also known as RNase L inhibitor (RLI) or 2'-5'-oligoadenylate-binding protein, is a member of the ATP-binding cassette (ABC) transporter family with a unique role in translation and ribosome recycling.
- Functions as a nucleoside-triphosphatase (NTPase) that hydrolyzes ATP, GTP, UTP, and CTP, and facilitates ribosome disassembly by splitting 80S ribosomes into free 60S and 40S subunits. Related references: PMID:20122402 PMID:21448132
- Essential for the No-Go Decay (NGD) pathway, where it is recruited to stalled ribosomes by the Pelota-HBS1L complex and drives disassembly, leading to mRNA degradation. Related references: PMID:21448132
- Acts as a negative regulator of the interferon-regulated 2-5A/RNase L pathway by inhibiting RNASEL activity and antagonizing 2-5A binding, thus modulating antiviral responses. Related references: PMID:9660177 PMID:11585831 PMID:9847332
- Localizes to both the cytoplasm and mitochondria; involved in mitochondrial quality control through PINK1-regulated ubiquitination and mitophagy. Related references: PMID:29861391
- Contains iron-sulfur clusters (4Fe-4S) and ATP-binding domains, and is subject to post-translational modifications including phosphorylation and ubiquitination.
- During HIV-1 infection, may act as a chaperone for capsid assembly, and during EMCV and HIV-1 infections, plays a role in down-regulating the 2-5A/RNASEL pathway.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 500-599 of human ABCE1; the antibody may preferentially recognize a linear epitope, which can be advantageous for Western blotting but may require optimization in immunofluorescence where native conformation is preserved.
- Validated for Western blotting (WB), immunofluorescence/immunocytochemistry (IF/ICC), and ELISA; for best results, titrate the antibody in your specific experimental system and include appropriate controls.
- The target protein has a predicted molecular weight of ~67 kDa; consider using a mid-percentage SDS-PAGE gel and standard PVDF or nitrocellulose membranes for transfer.
- Cross-reactivity with human and mouse ensures suitability for studies in both species, but verify specificity in your model organism through knockdown or knockout controls if necessary.
