Biological Background: ABCD1 Function and Localization
- ABCD1, also known as adrenoleukodystrophy protein (ALDP), is an ATP-dependent transporter of the ATP-binding cassette (ABC) family that mediates the transport of very long chain fatty acid (VLCFA)-CoA from the cytosol into the peroxisome lumen (PubMed:11248239, PubMed:15682271, PubMed:16946495). - The protein also possesses fatty acyl-CoA thioesterase (ACOT) activity, hydrolyzing VLCFA-CoA into free VLCFA prior to ATP-dependent translocation, a step essential for peroxisomal VLCFA import (PubMed:29397936, PubMed:33500543). Related references: PMID:29397936, PMID:33500543

Experimental Guidance and Technical Tips
- The immunogen spans amino acids 650–745 of the C-terminal region of human ABCD1, which is expected to be exposed on the cytoplasmic side of peroxisomal and other organelle membranes.
- For Western blotting, a band near 83 kDa is predicted; we recommend using positive control lysates from human or mouse tissues/cell lines known to express ABCD1 (e.g., adrenal gland, brain, or fibroblasts).
- The antibody’s human–mouse cross-reactivity enables its use in heterologous expression or knock-in models; however, expression levels and post‑translational modifications should be verified in the chosen experimental system.
- For ELISA applications, consider titrating the antibody to determine optimal coating and detection conditions; pairing with a validated detection antibody may enhance sensitivity.
