Biological Background: ABCC5/MRP5 Function and Localization
- ABCC5/MRP5 is a member of the ATP-binding cassette (ABC) transporter subfamily C, also designated multidrug resistance-associated protein 5, MOAT-C, SMRP, and pABC11.
- Acts as an ATP-dependent transporter extruding endogenous metabolites (cAMP, cGMP, folic acid, N-lactoyl-amino acids) and xenobiotics. Related references: PMID:10893247, PMID:12637526, PMID:12695538
- Functions as a general glutamate conjugate and analog transporter, limiting brain levels of endogenous metabolites, drugs, and toxins. Related references: PMID:26515061
- Confers resistance to the antiviral agent PMEA. Related references: PMID:12695538
- Transports anticancer drugs like methotrexate and nucleotide analogs with low affinity; its exact role in drug resistance remains to be fully elucidated. Related references: PMID:10840050, PMID:12435799, PMID:12695538
- Acts as a heme transporter required for translocation of cytosolic heme to the secretory pathway. Related references: PMID:24836561
- May regulate energy metabolism by modulating GLP-1 secretion from enteroendocrine cells (by similarity).
- Subcellular localization includes basolateral and apical cell membranes, Golgi apparatus lumen, endosome membrane, and cytoplasmic granules.
- Ubiquitously expressed with high levels in brain and muscle. Related references: PMID:10438534, PMID:15501592, PMID:9827529
- Predominant isoform in retinal pigment epithelium, bladder, and stomach.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 1101–1200 of human ABCC5 (NP_005679.2), located in the C-terminal region; this design may allow recognition of multiple splice variants.
- For immunofluorescence (IF-P), consider optimizing fixation and permeabilization conditions to detect membrane-localized ABCC5, as the protein resides in cell membranes and intracellular compartments.
- In ELISA applications, use the immunizing peptide as a positive control to confirm antibody reactivity, and note that cross-reactivity with other species has not been evaluated.
