Biological Background: ABCB8 Function and Localization
- ABCB8, also known as M-ABC1 or MITOSUR, is the ATP-binding subunit of the mitochondrial ATP-sensitive potassium channel (mitoK(ATP)).
Related references: PMID:31435016 - Together with the pore-forming subunit CCDC51/MITOK, ABCB8 mediates ATP-dependent potassium currents across the mitochondrial inner membrane; channel activity is inhibited by increased intracellular ATP.
Related references: PMID:31435016 - ABCB8 is involved in mitochondrial iron transport, critical for cellular iron homeostasis and iron-sulfur cluster biogenesis.
Related references: PMID:30623799 - The protein is required for normal cardiac function, likely by influencing mitochondrial iron export and maturation of cytosolic iron-sulfur cluster-containing enzymes (By similarity).
- ABCB8 is a member of the ATP-binding cassette (ABC) transporter superfamily and localizes to the mitochondrial inner membrane.
- Ubiquitously expressed in human tissues, it contains nucleotide-binding domains essential for ATP-dependent transport.
- Alternative splicing generates multiple isoforms of ABCB8, contributing to functional diversity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 469-718 of human ABCB8, a region within the nucleotide-binding domain; this design targets a conserved functional domain, supporting cross-reactivity with mouse and rat.
- For Western blotting, a band at approximately 80 kDa is expected in lysates from human, mouse, or rat tissues; validate loading controls and consider mitochondrial enrichment.
- In immunohistochemistry (IHC) and immunofluorescence (IF/ICC), anticipate mitochondrial staining patterns; optimization of antigen retrieval and permeabilization may be necessary.
- For ELISA, the polyclonal nature may provide broad epitope recognition; include appropriate controls for specificity.
