Biological Background: ABCB7 Function and Localization
- ABCB7, also known as ABC7 or Atm1p, is a member of the ATP-binding cassette (ABC) transporter family, specifically the B subfamily, encoded by the human ABCB7 gene.
- It functions as a mitochondrial inner membrane transporter that exports glutathione-coordinated iron-sulfur clusters, such as [2Fe-2S]-(GS)4, in an ATP-dependent manner, which is essential for the maturation of cytosolic Fe/S cluster-containing proteins. Related references: PMID:10196363 PMID:17192393 PMID:33157103
- Through a functional complex with ferrochelatase (FECH) and ABCB10, ABCB7 also plays a role in cellular iron homeostasis, mitochondrial function, and heme biosynthesis. Related references: PMID:30765471
- In cardiomyocytes, ABCB7 regulates cellular iron levels and reactive oxygen species (ROS) via interaction with COX4I1.
- ABCB7 may contribute to hematopoiesis.
- The protein is localized to the mitochondrial inner membrane and is subject to post-translational modifications such as acetylation and phosphorylation.
- Human ABCB7 gene is associated with disease variants, underscoring its clinical relevance.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering the C-terminal region (aa 503–753) of human ABCB7, which may confer epitope specificity. Consider this when designing experiments with truncated or isoform variants.
- For Western blotting, the predicted molecular weight is approximately 83 kDa. Use appropriate positive controls (e.g., tissues or cell lines from human, mouse, or rat) and ensure thorough mitochondrial lysis under denaturing conditions.
- The antibody is reported for WB, IHC-P, IF/ICC, and ELISA. Dilutions should be optimized in the relevant sample system; a starting point can be determined through serial dilution testing.
- When performing immunohistochemistry or immunofluorescence, note the mitochondrial inner membrane localization of ABCB7; antigen retrieval and permeabilization steps may be required for optimal signal.
