Biological Background: ABCB11 (BSEP) Function and Localization
- ABCB11, also known as Bile salt export pump (BSEP), is an ATP-binding cassette (ABC) transporter that mediates the ATP-dependent transport of major hydrophobic bile salts across the canalicular membrane of hepatocytes, thereby playing a crucial role in hepatic bile acid homeostasis and biliary lipid secretion. - It preferentially transports taurine-conjugated bile salts over glycine-conjugated species, and also handles non-bile acid substrates such as pravastatin and fexofenadine in an ATP-dependent manner. Related references: PMID:16332456, PMID:15901796, PMID:18245269
- Encoded by the ABCB11 gene, this protein belongs to the MDR/TAP subfamily of ABC transporters and is essential for enterohepatic circulation of bile acids.
- Subcellularly, BSEP localizes to the apical (canalicular) membrane of hepatocytes, and also cycles through recycling endosomes and endosomal compartments, as indicated by its presence in the apical cell membrane, recycling endosome membrane, and endosomes.

- For Western blot (WB), a specific band at approximately 146 kDa is expected; validate detection with positive control lysates from liver or ABCB11-overexpressing cells.
- In immunofluorescence (IF/ICC), permeabilization is required, and a canalicular staining pattern should be observed in polarized hepatocyte models; co-staining with canalicular markers is recommended.
- For ELISA applications, consider using the antibody for capture or detection after confirming binding affinity and specificity with recombinant ABCB11 fragments or purified protein.
