Biological Background: ABCB11 Function and Localization
- ABCB11, also known as Bile Salt Export Pump (BSEP), is a transmembrane transporter belonging to the ATP-binding cassette (ABC) sub-family B member 11.
- Functions as the major canalicular bile salt transporter, catalyzing ATP-dependent export of taurine- and glycine-conjugated cholic acid across the hepatocyte apical membrane, crucial for bile acid homeostasis.
Related references: PMID:15791618, PMID:16332456, PMID:18985798 - Exhibits higher transport efficiency for taurine-conjugated bile salts compared to glycine-conjugated counterparts.
Related references: PMID:16332456 - Additionally transports non-bile acid compounds such as pravastatin and fexofenadine, indicating a role in biliary drug excretion.
Related references: PMID:15901796, PMID:18245269 - Predominantly expressed in the liver, localized to the canalicular microvilli and subcanalicular vesicles of hepatocytes.
- Subcellularly cycles between the apical membrane and recycling endosomes, with endosome-to-membrane trafficking essential for its function.
- Genetic mutations in ABCB11 are associated with progressive familial intrahepatic cholestasis type 2 (PFIC2) and other cholestatic diseases.
- Post-translationally modified by phosphorylation and ubiquitination, which may regulate its activity and stability.
Experimental Guidance and Technical Tips
- Because the immunogen is a peptide from the C-terminal region (aa 1050-1150), this antibody may recognize the full-length protein but not necessarily isoforms lacking this epitope.
- For IF/ICC, consider using appropriate permeabilization and blocking protocols for membrane proteins; validate staining in liver tissue or hepatocyte cell lines.
- In ELISA, the antibody may serve as a capture or detection reagent; optimize coating antigen and detection conditions.
- Since the protein is large (146 kDa), ensure adequate protein transfer and handling in any downstream applications.
- Cross-reactivity with mouse and rat indicates potential for use in rodent models; validate species-specific reactivity in the relevant sample system.
