Biological Background: ABCA8 Function and Localization
- Belongs to the ATP-binding cassette (ABC) transporter family, subfamily A, member 8; functions as an organic anion transporter and cholesterol efflux pump.
- Catalyzes ATP-dependent import of organic anions taurocholate, estrone sulfate, and ochratoxin A; also mediates cholesterol efflux independent of apolipoprotein and contributes to sphingomyelin production in oligodendrocytes. Related references: PMID:12379217, PMID:23560799
- Mediates ATP-dependent efflux of cholesterol and taurocholate; interaction with ABCA1 potentiates cholesterol efflux to lipid-free APOA1, thereby regulating high-density lipoprotein levels. Related references: PMID:29300488, PMID:28882873
- Localizes primarily to the cell membrane and basolateral cell membrane, consistent with its transport functions.
- Widely expressed in human tissues, with higher expression in heart, skeletal muscle, liver, and white matter regions (superior frontal and inferior temporal). Related references: PMID:12379217, PMID:28882873, PMID:29300488
- Contains ATP-binding domains, multiple transmembrane helices, and is classified as a glycoprotein and alternative splicing variant.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a cytoplasmic region (aa 425–495); consider using appropriate membrane protein extraction methods for optimal detection.
- For Western blot, a high molecular weight band of ~184 kDa is expected; use a suitable gel percentage and transfer conditions.
- When performing ELISA, validate coating conditions and antibody dilution in your specific assay system using known positive controls (e.g., heart or liver lysates).
- Cross-reactivity with rat is indicated; verify reactivity in rat samples and optimize protocols accordingly.
