Biological Background: ABCA7 Function and Localization
- ABCA7 is a phospholipid-transporting ATPase that catalyzes the translocation of phosphatidylserine and, to a lesser extent, phosphatidylcholine across membranes, playing a critical role in lipid homeostasis. Related references: PMID:24097981
- It functions in apolipoprotein-mediated lipid efflux, binding APOA1 and participating in cholesterol and phospholipid efflux from cells, which is important for reverse cholesterol transport. Related references: PMID:12917409 PMID:14570867 PMID:14592415
- ABCA7 is involved in macrophage-mediated phagocytosis, facilitating the clearance of apoptotic cells and amyloid-beta, and its phagocytic activity is stimulated by APOA1 or APOA2. Related references: PMID:12925201
- The protein regulates cellular ceramide homeostasis, particularly during keratinocyte differentiation, highlighting its role in sphingolipid metabolism. Related references: PMID:12925201
- ABCA7 influences amyloid-beta production and clearance; it limits amyloid-beta generation by regulating amyloid precursor protein (APP) endocytosis/processing and promotes amyloid-beta phagocytosis by microglia and macrophages, linking it to Alzheimer's disease pathology. Related references: PMID:26260791
- Localized to the cell membrane, Golgi apparatus, early endosomes, endoplasmic reticulum, and phagocytic cup, ABCA7's subcellular distribution supports its roles in lipid transport and phagocytosis.
- Two major isoforms exist: isoform 1 is strongly expressed in brain and bone marrow, while isoform 2 is abundant in lymph node, spleen, thymus, and trachea, suggesting tissue-specific functions.
- The protein is widely expressed, with high levels in myelo-lymphatic tissues (peripheral leukocytes, thymus, spleen, bone marrow) and detectable expression in the hippocampus and cerebellum, consistent with its immune and neural roles.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 2010–2146) of human ABCA7; verify sequence conservation if detecting both isoforms or cross-reacting species.
- For Western blot (WB), note the large predicted molecular weight (~234 kDa); use low-percentage gels, extended transfer times, and high molecular weight markers.
- In immunofluorescence (IF/ICC), expect plasma membrane, Golgi, endosomal, and phagocytic cup staining; co-localization with organelle markers can confirm specificity.
- For ELISA, consider using the antibody as a capture or detection reagent; validate with recombinant ABCA7 protein or lysates from expressing cells.
- When testing mouse or rat samples, confirm cross-reactivity by aligning the immunogen sequence with the respective orthologs; the antibody is reported to recognize both species.
