Biological Background: ABCA4 Function and Localization
- ABCA4 (ATP-binding cassette sub-family A member 4) is also known as RIM ABC transporter, RmP, retinal-specific phospholipid-transporting ATPase, and Stargardt disease protein. Encoded by the ABCA4 gene, it is a transmembrane protein with a molecular weight of approximately 256 kDa.
- Catalyzes the ATP-dependent transport of retinal-phosphatidylethanolamine conjugates (11-cis and all-trans N-retinylidene-PE) from the lumen to the cytoplasmic leaflet of photoreceptor outer segment disk membranes. This flippase activity clears photogenerated all-trans-retinal, preventing accumulation and toxic bisretinoid formation (PubMed:10075733, PubMed:20404325, PubMed:22735453).
Related references: PMID:10075733, PMID:20404325, PMID:22735453 - Displays intrinsic ATPase activity in vitro, and may exhibit GTPase activity modulated by the lipid environment and retinoid compounds (PubMed:33605212, PubMed:39128720, PubMed:29847635, PubMed:22735453).
Related references: PMID:33605212, PMID:39128720, PMID:29847635, PMID:22735453 - Localizes to the membrane of photoreceptor outer segment disks, and has also been detected in the endoplasmic reticulum and cytoplasmic vesicles.
- Tissue expression is retinal-specific, predominantly found in the rim region of rod photoreceptor cells.
- Genetic variants in ABCA4 are linked to several inherited retinal dystrophies, including Stargardt disease (STGD1), cone-rod dystrophy (CORD3), retinitis pigmentosa (RP19), and age-related macular degeneration (ARMD2). The protein undergoes post-translational modifications such as glycosylation and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 110–300, an N-terminal region, which may provide reactivity with various ABCA4 isoforms or fragments. Confirm specificity in your model system.
- For Western blotting, note the high molecular weight (~256 kDa). Use a low-percentage SDS-PAGE gel (e.g., 6–8%) and optimize transfer conditions (e.g., overnight wet transfer) for efficient blotting of large proteins.
- For ELISA applications, this polyclonal antibody can be used as a capture or detection reagent. Pair with a matched monoclonal or another polyclonal antibody targeting a distinct epitope for sandwich assays, and validate performance with known positive controls.
- Recommended dilution factors should be determined empirically; literature and general guidelines can serve as starting points.
