Experimental Guidance and Technical Tips
- Given the immunogen corresponds to the C-terminal region (aa 1870–2120) of human ABCA1, the antibody may recognize epitopes in this distal domain, which is predicted to be extracellular; thus, it may be suitable for detecting surface-localized ABCA1 in IF/ICC under non-permeabilizing conditions. However, validate in the relevant sample system.
- Due to the large molecular weight of ABCA1 (~254 kDa), consider extended transfer times or low-percentage gels (e.g., 6–8% SDS-PAGE) and verify transfer efficiency with a protein ladder or total protein stain.
- For WB, a loading control of similar high molecular weight may be necessary; alternatively, Ponceau S staining can confirm transfer.
- Cross-reactivity with mouse and rat has been reported; however, the degree of reactivity in these species should be empirically determined using tissue or cell lysates expressing ABCA1.
