Biological Background: ABCA1 Function and Localization

  • ABCA1 (ATP-binding cassette sub-family A member 1), also known as ABC-1, CERP, and cholesterol efflux regulatory protein, is a member of the ATP-binding cassette (ABC) transporter superfamily.
  • It functions as a phospholipid-transporting ATPase, translocating phosphatidylcholine and phosphatidylserine from the cytoplasmic to the extracellular leaflet of membranes, coupled to ATP hydrolysis. This activity is essential for the transfer of phospholipids to apolipoproteins, initiating the formation of nascent high-density lipoproteins (HDLs). Related references: PMID:24097981, PMID:35974019
  • ABCA1 also mediates the efflux of intracellular cholesterol to apolipoproteins, a critical step in reverse cholesterol transport and HDL maturation. Related references: PMID:10533863, PMID:14754908, PMID:24097981
  • Localized primarily at the cell membrane and in endosomes, ABCA1 is widely expressed but most abundant in macrophages, reflecting its key role in macrophage cholesterol homeostasis.
  • Post-translational modifications include phosphorylation (phosphoprotein) and palmitoylation (lipidation), and the protein contains disulfide bonds and glycosylation sites, which may regulate its stability and activity.
  • Given its function in lipid metabolism, ABCA1 is implicated in atherosclerosis and related cardiovascular diseases; mutations in ABCA1 are associated with Tangier disease and familial HDL deficiency.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 1170-1350 within the C-terminal region of human ABCA1. This region is predicted to be intracellular, and antibodies raised against it may detect full-length ABCA1 in denaturing Western blots. Consider validating specificity in cell lines with known ABCA1 expression, such as macrophages or hepatocytes.
  • For Western blotting, note the large predicted molecular weight of ABCA1 (~254 kDa). Use low-percentage polyacrylamide gels (e.g., 6% gel) and extended transfer times to ensure efficient transfer of high-molecular-weight proteins. Include appropriate molecular weight markers.
  • This antibody is reported to cross-react with mouse and rat ABCA1. When using in these species, verify reactivity by testing a positive control, such as macrophage-derived cells or tissues known to express ABCA1.
  • For ELISA applications, optimize the antibody concentration as a capture or detection reagent. The antibody’s reactivity in ELISA may allow for quantitative measurement of ABCA1 in biological samples; however, pairing with a suitable detection antibody may be necessary for sandwich formats.
