Biological Background: ABCA1 Function and Localization

  • ABCA1 (ATP-binding cassette sub-family A member 1), also known as ABC-1 or cholesterol efflux regulatory protein, is a phospholipid-transporting ATPase.
  • It catalyzes the ATP-dependent translocation of phospholipids, preferentially phosphatidylcholine, from the cytoplasmic to the extracellular/lumenal leaflet of the membrane. Related references: PMID:24097981 PMID:35974019
  • This translocase activity mediates phospholipid and cholesterol transfer to apolipoproteins, driving the formation of nascent high-density lipoproteins (HDL). Related references: PMID:14754908 PMID:10533863 PMID:24097981
  • Mechanistically, ABCA1 forces phospholipids through a membrane gateway and annulus into an elongated hydrophobic tunnel in its extracellular domain. Related references: PMID:35974019
  • The protein localizes to the cell membrane and endosomes.
  • ABCA1 is widely expressed, with highest abundance in macrophages, consistent with its role in cholesterol homeostasis.
  • It undergoes post-translational modifications including disulfide bond formation, glycosylation, phosphorylation, and palmitoylation.
  • Due to its critical function in reverse cholesterol transport, ABCA1 is implicated in atherosclerosis and HDL deficiency disorders.

Experimental Guidance and Technical Tips

  • The immunogen sequence (amino acids 1170–1350) resides in the C-terminal region of human ABCA1. This region shows high homology with mouse and rat orthologs, supporting the reported cross-reactivity.
  • For Western blotting, the predicted molecular weight is ~254 kDa. Use low-percentage SDS-PAGE gels (e.g., 6–8%) and optimized transfer conditions for high-molecular-weight proteins.
  • Consider using macrophage cell lysates (e.g., THP-1, RAW 264.7) as positive controls to validate detection of endogenous ABCA1.
  • For ELISA applications, test both direct and sandwich formats to determine optimal sensitivity with this monoclonal antibody.
  • Always include appropriate controls and validate performance in your specific experimental system.
