Biological Background: AANAT Function and Localization
- Serotonin N-acetyltransferase (AANAT), also known as aralkylamine N-acetyltransferase (AA-NAT) or SNAT, is encoded by the AANAT gene.
- It catalyzes the N-acetylation of serotonin into N-acetylserotonin, the penultimate step in melatonin biosynthesis, thereby controlling the night/day rhythm of melatonin production in the pineal gland.
- AANAT belongs to the acyltransferase and transferase families and plays a key role in biological rhythms.
- The enzyme is primarily localized in the cytoplasm.
- It is highly expressed in the pineal gland and, at lower levels, in the retina, with weak expression in several brain regions and the pituitary gland.
- The protein undergoes phosphorylation and alternative splicing, and its three-dimensional structure has been determined.
- The molecular weight is approximately 23.3 kDa (UniProt), consistent with the observed band at 23 kDa.
