- AADAT (Kynurenine/alpha-aminoadipate aminotransferase, mitochondrial) is a pyridoxal phosphate-dependent aminotransferase encoded by the AADAT gene, also known as KAT2 or KYAT2.
- The enzyme exhibits broad substrate specificity, catalyzing transamination of aminoadipate, kynurenine, methionine, and glutamate, with additional activity toward tryptophan, aspartate, and hydroxykynurenine.
- It utilizes various oxo-acid amino-group acceptors, preferring 2-oxoglutarate, 2-oxocaproic acid, phenylpyruvate, and alpha-oxo-gamma-methiol butyric acid, and can employ glyoxylate in vitro.
- AADAT localizes primarily to the mitochondrion, consistent with its role in amino acid metabolism.
- Expression is highest in liver, with detectable levels in heart, brain, kidney, pancreas, prostate, testis, and ovary.
- The protein undergoes alternative splicing and acetylation, and its 3D structure has been determined.
