- AADAT, also known as kynurenine/alpha-aminoadipate aminotransferase mitochondrial, is encoded by the AADAT gene and alternatively named KAT2, KYAT2. It is a pyridoxal phosphate-dependent aminotransferase.
- The enzyme exhibits broad substrate specificity, catalyzing transamination of aminoadipate, kynurenine, methionine, and glutamate. It also acts on tryptophan, aspartate, and hydroxykynurenine, accepting various oxo-acids with a preference for 2-oxoglutarate.
- AADAT plays a key role in amino acid metabolism, converting kynurenine to kynurenic acid, a neuroprotective metabolite, linking it to neurodegenerative disease research.
- Subcellularly, AADAT is localized to the mitochondrion, consistent with its role in mitochondrial amino acid degradation and energy metabolism.
- Tissue expression is highest in liver, with detectable levels in heart, brain, kidney, pancreas, prostate, testis, and ovary, suggesting widespread metabolic functions.
