Biological Background: Alpha-synuclein Function and Localization
- Alpha-synuclein, encoded by SNCA, is a 140-amino acid protein also known as NACP (Non-A beta component of AD amyloid) or PARK1/4, predominantly expressed in presynaptic terminals of the central nervous system.
- It regulates synaptic vesicle trafficking and neurotransmitter release by enhancing vesicle priming, fusion, and dilation of exocytotic fusion pores, partly through increasing local Ca²⁺ release (PubMed:28288128, PubMed:30404828). Related references: PMID:28288128 PMID:30404828
- In its membrane-bound multimeric state, alpha-synuclein acts as a molecular chaperone for SNARE complex assembly, working with cysteine string protein-alpha to sustain normal exocytosis, particularly important during aging (PubMed:20798282). Related references: PMID:20798282
- It modulates dopamine neurotransmission by interacting with the dopamine transporter (DAT1) (PubMed:26442590). Related references: PMID:26442590
- Subcellularly, alpha-synuclein localizes to the cytoplasm, membrane, nucleus, synapses, and can be secreted, with enrichment in cell projections like axons.
- The protein undergoes post-translational modifications including acetylation, phosphorylation, and ubiquitination, binds copper, and its aggregation is central to Parkinson disease and other synucleinopathies.
Experimental Guidance and Technical Tips
- The immunogen covering the full-length protein (aa 1-140) ensures recognition of the native conformation in various applications. Consider antigen retrieval for IF-P and optimize dilution for IP.
- For WB, validate detection in relevant human, mouse, or rat brain tissue lysates given the reported cross-reactivity. The predicted molecular weight is ~14 kDa, but aggregation states may appear as higher bands.
- Use the antibody in validated applications (WB, IP, IF/ICC, IF-P, ELISA) and confirm performance in your specific experimental system.
