Biological Background: α-Synuclein Function and Localization

  • Alpha-synuclein (SNCA) is a small presynaptic protein belonging to the synuclein family, encoded by the SNCA gene (also known as NACP, PARK1/4). It is highly expressed in brain, particularly at presynaptic terminals.

  • For Western blotting, a band at approximately 14 kDa is expected. Optimize sample preparation (e.g., avoiding boiling, using mild detergents) to preserve native aggregation states if studying oligomers.
  • In IF/ICC and IF‑P, validate the antibody in neuronal cell lines or brain tissue sections known to express α‑synuclein; co‑staining with synaptic markers may help confirm localization.
  • ELISA protocols should include appropriate controls and consider the assay format (direct, sandwich) to detect α‑synuclein in biological fluids, keeping in mind the protein’s tendency to aggregate.
