- 5T4, also known as trophoblast glycoprotein, M6P1, 5T4AG, or WAIF1, is encoded by the TPBG gene and belongs to the leucine-rich repeat protein family.
- It functions as an inhibitor of Wnt/beta-catenin signaling by indirectly interacting with LRP6 and blocking Wnt3a-dependent LRP6 internalization.
- The protein localizes to the cell membrane and contains a signal peptide, a large extracellular domain with leucine-rich repeats, and a single transmembrane helix.
- 5T4 undergoes post-translational modifications including glycosylation and phosphorylation, and contains disulfide bonds, contributing to its structural stability.
- In normal tissues, 5T4 is highly expressed in trophoblasts during early development (from 9 weeks) and is restricted to a few epithelial cell types in adults.
- It is frequently overexpressed in a variety of carcinomas, making it a recognized oncofetal antigen with potential as a diagnostic and therapeutic target.
