Biological Background: TP53-binding protein 1 Function and Localization

  • 53BP1, also known as p202, TDRD30, or p53BP1, is a key double-strand break (DSB) repair protein involved in DNA damage response, telomere dynamics, and class-switch recombination (CSR) during antibody genesis. Related references: PMID:12364621 PMID:17190600 PMID:21144835

  • It promotes non-homologous end joining (NHEJ)-mediated repair of DSBs and specifically counteracts BRCA1-dependent homologous recombination (HR) repair. Related references: PMID:22553214 PMID:23333306 PMID:23727112

  • Upon DNA damage, ATM-mediated phosphorylation of 53BP1 promotes its interaction with RIF1 and dissociation from TIRR, facilitating recruitment to DSB sites. Related references: PMID:28241136

  • 53BP1 is recruited to DSBs by recognizing histone H2A monoubiquitinated at K15 (H2AK15Ub) and histone H4 dimethylated at K20 (H4K20me2). Related references: PMID:17190600 PMID:23760478 PMID:27153538

  • It is required for immunoglobulin class-switch recombination (CSR), a process that generates DNA DSBs. Related references: PMID:23345425

  • 53BP1 also promotes NHEJ at dysfunctional telomeres through interaction with PAXIP1. Related references: PMID:23727112

  • Subcellularly, 53BP1 localizes to the nucleus, chromosomes, and specifically to centromeres and kinetochores.

  • Alternative splicing produces multiple 53BP1 isoforms, contributing to functional diversity in DNA repair.

Experimental Guidance and Technical Tips

  • As a polyclonal antibody raised against a recombinant fragment (aa 895-1031), it is expected to recognize the full-length 53BP1 protein (~214 kDa) and possibly degradation products in western blotting. Optimize exposure and loading controls.
  • For WB, a typical starting dilution range of 1:500–1:2000 is often used for polyclonal antibodies; however, the optimal working concentration must be determined empirically in your sample system.
  • In IHC-P, antigen retrieval may be required due to the nuclear localization of 53BP1; validate staining specificity with appropriate tissue controls and consider using a nuclear counterstain.
  • The antibody shows cross-reactivity with mouse and rat 53BP1, as confirmed by the supplier, making it useful for translational studies; however, sequence alignment of the immunogen region across species should be considered.
  • Since 53BP1 is a phosphoprotein involved in DNA damage signaling, treatment of cells with DNA-damaging agents (e.g., ionizing radiation, etoposide) may enhance detection of foci formation in immunofluorescence or modulate signal in WB.
  • For ELISA, the antibody can be used to capture or detect recombinant 53BP1 fragments or cell lysates; appropriate blocking and washing conditions should be optimized.
