Biological Background: 15-PGDH/HPGD Function and Localization
- 15-PGDH (HPGD/SDR36C1) is a member of the short-chain dehydrogenase/reductase family that catalyzes NAD-dependent oxidation of hydroxylated polyunsaturated fatty acids, including prostaglandins, lipoxins, and resolvins, producing their corresponding keto metabolites. Related references: PMID:10837478, PMID:16757471, PMID:16828555
- By degrading pro-proliferative prostaglandin E2, it acts as a tumor suppressor; its oxo-fatty acid products can abrogate pro-inflammatory cytokine expression. Related references: PMID:15574495, PMID:25586183
- It converts resolvins E1, D1, and D2 to oxo metabolites, thereby inactivating these resolution-phase mediators, which impacts acute inflammation and obesity-induced adipose inflammation. Related references: PMID:16757471, PMID:22844113
- Subcellularly localized to the cytoplasm.
- Detected at the protein level in colon epithelium.
- Keywords: Alternative splicing, Disease variant, Fatty acid metabolism, Lipid metabolism, NAD, Oxidoreductase, Prostaglandin metabolism, Tumor suppressor.
Experimental Guidance and Technical Tips
- The immunogen spans the full-length human 15-PGDH (amino acids 1-266), suggesting recognition of multiple epitopes; perform appropriate validation in the relevant sample system.
- For Western blotting, a band of approximately 29 kDa is expected; include positive and negative controls and consider using reducing conditions.
- In ELISA applications, optimize coating antigen and detection conditions; this polyclonal antibody may be suitable for capture or detection formats depending on assay design.
- Cross-reactivity with mouse and rat has been observed; verify performance and specificity in these species using appropriate tissue or cell lysates.
