Biological Background: 12 Lipoxygenase/ALOX12 Function and Localization

  • ALOX12 is a polyunsaturated fatty acid lipoxygenase, also known as 12S-lipoxygenase or platelet-type lipoxygenase, encoded by the ALOX12 gene (Gene ID: 239).
  • It catalyzes the regio- and stereo-specific oxygenation of free and esterified polyunsaturated fatty acids to generate lipid hydroperoxides, with arachidonate as a major substrate yielding (12S)-HPETE. Related references: PMID:17493578, PMID:18311922, PMID:1851637
  • Through (12S)-HPETE production, it regulates platelet activation. Related references: PMID:8319693, PMID:8500694
  • It also catalyzes the epoxidation of double bonds, e.g., converting (14S)-HPDHA to (13S,14S)-epoxy-DHA, and participates in the biosynthesis of specialized pro-resolving mediators like resolvin D5. Related references: PMID:23504711, PMID:32404334
  • ALOX12 transforms leukotriene A4 into the vasoactive lipoxins LXA4 and LXB4, modulating neutrophil function. Related references: PMID:8250832
  • It is considered protumorigenic, promoting VEGF expression and integrin beta-1 to influence tumor survival and metastasis. Related references: PMID:16638750, PMID:22237009, PMID:9751607
  • ALOX12 also plays a role in preventing apoptosis in vascular smooth muscle cells. Related references: PMID:23578768
  • Subcellular localization includes cytoplasm, cytosol, and membrane; tissue expression is noted in vascular smooth muscle cells.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a C‑terminal region (aa 483‑663) of human ALOX12; consider sequence alignment to verify epitope conservation in cross‑reactive species (mouse, rat).
  • For Western blot, a band at approximately 76 kDa is expected; validate with appropriate positive controls such as human platelet lysates or vascular smooth muscle cells.
  • In immunofluorescence (IF‑P), fixation and permeabilization conditions may influence detection; optimize for your cell type and consider inclusion of organelle markers to assess localization.
  • For ELISA, the antibody may be used for capture or detection; pair with appropriate standards and perform cross‑reactivity testing.
