Biological Background: ANGEL2 Function and Localization
-
ANGEL2 (also known as Ccr4d or KIAA0759L) is an RNA 2',3'-cyclic phosphatase that specifically hydrolyzes the 2',3'-cyclic phosphate at the 3' end of RNA, generating a linear 3'-phosphate group.
Related references: PMID:32732418 PMID:36180430 -
It plays a critical role in mitochondrial RNA maturation and processing, particularly for non-canonically generated transcripts.
-
ANGEL2 antagonizes pre-tRNA processing and XBP1 mRNA splicing during the unfolded protein response by limiting 2',3'-cyclic phosphate substrates for the tRNA ligase complex.
Related references: PMID:32732418 -
The protein promotes tRNA recycling by removing 2',3'-cyclic phosphate groups from cleaved tRNAs, enabling TRNT1 to reattach the CCA tail essential for aminoacylation and translation.

Subcellularly, ANGEL2 localizes to the mitochondrial matrix.
-
The gene undergoes alternative splicing, and the enzyme requires magnesium as a cofactor; it is classified as a hydrolase involved in both mRNA and tRNA processing.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 30–170 in the N-terminal region of human ANGEL2. Consider verifying antibody recognition of endogenous full-length protein or possible isoforms using recombinant protein controls.
- For Western blot, the predicted molecular weight of ANGEL2 is approximately 62 kDa. Optimize blocking and detection conditions to minimize non-specific binding common with polyclonal preparations.
- In ELISA applications, consider including peptide competition or reference standards to confirm specificity and sensitivity.
- Because ANGEL2 resides in the mitochondrial matrix, ensure appropriate lysis and sample preparation protocols to effectively solubilize this organellar protein from cells or tissues.
