Biological Background: ALDH9A1 Function and Localization
- ALDH9A1, also known as 4-trimethylaminobutyraldehyde dehydrogenase, is a member of the aldehyde dehydrogenase family and functions as an NAD-dependent oxidoreductase.
- It converts gamma-trimethylaminobutyraldehyde into gamma-butyrobetaine with high efficiency, and also catalyzes the oxidation of a broad range of aldehydes from biogenic amines and polyamines, though with lower efficiency.
- The enzyme localizes predominantly in the cytoplasm and cytosol.
- ALDH9A1 is detected in brain at the protein level. Related references: PMID:8645224
- ALDH9A1 exhibits high expression in adult liver, skeletal muscle, and kidney, with lower levels in heart, pancreas, lung, and brain. Related references: PMID:8786138
- Within the brain, ALDH9A1 is expressed in all regions, with highest levels in the spinal cord and lowest in the occipital pole. Related references: PMID:8786138
- The protein undergoes acetylation and alternative splicing, which may generate multiple isoforms.
- Keywords include 3D-structure, indicating experimentally determined structural data is available.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1–180) of human ALDH9A1, which may provide specificity for the target. Consider validating reactivity in the relevant sample system.
- For Western blot (WB), a band around 54 kDa is expected; optimize lysate preparation and antibody dilution.
- In immunofluorescence/immunocytochemistry (IF/ICC), appropriate fixation and permeabilization conditions should be tested to preserve antigenicity.
- For ELISA, the antibody may be suitable as a capture or detection reagent; pairing with a matched antibody is recommended.
- Cross-reactivity with human, mouse, and rat ALDH9A1 suggests the antibody can be used across these species, but verification in each species is advised.
